The success in isolating the voices from a song depends almost entirely on the way the song has been mixed and will vary considerably. There is no perfect solution, although many times the artifacts that remain can be minimized within a mix. Generally, you will get the best hit with the music in which the voices are moved to a specific position in the mix, usually in the center. Then you will need some EQ or low frequency spectral removal after completing the extraction step.

The tool you want to use in Audition is the Central channel Extractor effect, located under the Effects> Stereo Images menu. Start with the Acapella preset, then change the Extract parameter to Custom for additional control. Select the region you want to isolate (it is often better to isolate the passages separately, because, for example, the verses can be mixed very differently than a choir) and start playback with the effect open. Use the button to temporarily bypass the effect so you can see the differences.

Adjusts the Phase Angle and Pan parameters to maximize voices while minimizing instrumentation. Use frequency range parameters to reduce some of the low-end and high-end mixes that are not associated with the voice. The two vertical controls on the right let you adjust the levels of what is isolated relative to everything else. (Flip-Flop to create a Karaoke mix.) Below the Discrimination tab, the most important parameters will be the Crossover Bleed and Phase Discrimination controls. You can adjust the FFT parameters under Advanced tab, though I would not recommend it until you feel comfortable with the tool. You do not necessarily need to know exactly what the parameter ever does, but listen carefully while making adjustments and you will begin to understand how each parameter affects the result.
